Histological Changes of Lymph Nodes and Spleen during the "Ture" Primary Immune Response in Germ-free Colostrum-deprived Piglets.



Abstract
Germ-free colostrum-deprived piglets were obtained from disease-free Minnesota miniature sow. These animals were kept in the germ-free isolators during the period of experiment and randomly divided into four groups. Group A received a single injection of 1012 purified actinophage (MSP-2) intraperitoneally within five hours after the hysterectomy. Group B were uninjected littermate controls.
Each group was divided into two subgroups: one half of each group was kept germ-free and fed an antigen free diet, namely, sterile pyrogen-free distilled water for short term experiments(for 72 hours).
The other half of each group was kept germ-free and fed Mullsoy-diet for long term observation. These animals were sacrificed by bleeding at various intervals, and we took serum for antibody titration and took spleen and regional lymphnodes, that is, mesenteric, cervical, submaxillary, axillary, inguinal, and substernal lymph nodes for histological study. The paraffin sections were stained with H & E, M. G.P.(Methyl Green Pyronin) and silver. We examined them under the light microscope and had results as followings:
1. Germ-free colostrum-deprived piglets were free from detectable immuno-globulins in their sera and their tissues were free from "background" antibody forming cells. They were, therefore, immunologically "virgin".
